Loratinib/loratinib lorviqua is the third generation ALK inhibitor. As a follow-up treatment of crizotinib resistance, it can inhibit nine mutations of crizotinib resistance, and is especially suitable for the treatment of patients with advanced non-small cell lung cancer (NSCLC) resistant to other ALK.

Targeted drugs are suitable for the treatment of patients with non-small cell lung cancer NSCLC with ALK positive.
For patients with NSCLC who are ALK positive for the first time, the effective rate of this product is as high as 90%, and the intracranial effective rate is as high as 75%.
For ALK positive patients who had received crizotinib before, the effective rate of this product was 69%, and the intracranial effective rate reached 68%.
For patients with ALK positive and previously treated with ALK inhibitors other than crizotinib, the effective rate of this product was 33%, and the intracranial effective rate reached 42%.
For patients with ALK positive and previously treated with 2-3 ALK inhibitors, the effective rate of this product is 39%, and the intracranial effective rate is 48%.

The clinical results show that this product has clinical significance in the treatment of lung tumor and brain metastasis in advanced NSCLC patients with ALK positive and ROS1 positive.

[Notes for lorviqua of Loratinib/Lolatinib]
1. Brain (central nervous system) function problems. Many patients have brain function problems during the treatment with lorviqua, including thinking problems (such as forgetfulness or confusion), emotional problems (such as depression), speech problems, seeing or hearing unreal things (hallucinations) and seizures. In some patients, these problems are so serious that your health care provider may need you to stop taking lorviqua.
2. Increased levels of cholesterol and triglycerides (lipids) in the blood. During the treatment of lorviqua, the blood lipid level of most patients will increase.
3. If the blood lipid level in your blood increases during the treatment of lorviqua, your health care provider may need to start giving you drugs to reduce the blood lipid level.
4. If you are already taking a drug to lower the blood lipid level, your health care provider may need to increase your drug dosage.
5. Your health care provider should conduct blood tests before starting treatment, 1-2 months after starting treatment and during the treatment with lorviqua to check the blood lipid level.
6. Heart problems. Lorviqua may cause a very slow or abnormal heartbeat. Your healthcare provider should check your heart rate (electrocardiogram) before and during the treatment with lorviqua. If you feel dizzy, weak or abnormal heartbeat, please tell your health care personnel immediately. In some patients, these problems are so serious that your health care provider may need you to stop taking lorviqua or install a pacemaker.
7. Lung problems. Lorviqua may cause serious or life-threatening swelling (inflammation) of the lungs during treatment, which may lead to death. Symptoms may be similar to lung cancer.
8. If you have any new or worsening symptoms of lung problems, including dyspnea, shortness of breath, cough or fever, please tell your health care provider immediately.

The most common side effects include:
Swelling of arms, legs, hands and feet (edema)
Numbness and tingling sensation in joints or arms and legs (peripheral neuropathy)
